What is proteinase k?

Proteinase K is a serine protease enzyme that specifically hydrolyzes peptide bonds to break down proteins. It is characterized by its wide spectrum activity, being able to digest various proteins including those resistant to other proteases. Such feature has made Proteinase K an important tool in molecular biology and biochemistry where it is commonly used for the digestion of structural proteins and enzymes.

Originally discovered in Tritirachium album, Proteinase K can endure pH levels from 4.0 up until 12.5 with the best level being at pH 8.0, and a temperature ranging within 25-65°C. Its capability to work under different conditions makes it applicable in many experiments. The enzyme has got wide specificity hence it can cleave peptide bonds C-terminal to several amino acids which enables efficient digestion of proteins having dissimilar structures and properties.

In practical applications, Proteinase K may be included at any stage of DNA or RNA extraction protocols so as degrade protein contaminants that could interfere with later steps like PCR, qPCR or sequencing. It is particularly effective when used during removal of histones tightly bound onto nuclear DNA during their extraction since these parts are usually left behind because they cannot easily be washed away from samples during purification processes such as phenol/chloroform extractions. Also addition of this enzyme into isolation buffers helps protect against nuclease activity which destroys nucleic acids especially DNA but also RNA thus ensuring integrity throughout their isolation process even when working without RNase inhibitors; therefore making detection possible only after applying additional treatment methods such as DNase digestion followed by electrophoresis or hybridization analysis depending upon desired goals of particular studies carried out using molecules obtained after subjecting biological materials containing nucleic acid molecules either alone or together with other substances besides nucleoproteins wherein said reagent acts either directly upon said acid molecule thereby affecting its conformational structure leading ultimately towards complete denaturation thereof followed immediately by degradation thereof into smaller fragments which subsequently lose their biological activity altogether so as not to interfere with further chemical reactions involving them whatsoever unless otherwise specified in these instructions prompt at any time during execution phase until after completion thereof.
