What is poly a polymerase?

Posttranscriptional change of RNA particles in living beings is fundamentally performed by Poly(A) polymerase. The function of this enzyme is to append a long chain of adenosine nucleotides, also known as poly(A) tail, to the 3' end of messenger RNA (mRNA) molecules. Distinct from DNA sequence, the poly(A) tail is not templated but synthesized anew by PAP.

Polyadenylation is a necessary process for mRNA stability and efficient translation within the cytoplasm. As part of RNA processing, pre-mRNA undergoes several modifications like 5'-capping, splicing and 3'-cleavage after being transcribed by RNA polymerase II. Post cleaving at poly(A) site, PAP attaches a poly(A) tail to the newly formed 3’ terminus of mRNA molecule. Nuclear poly(A)-binding proteins (PABPNs) which regulate addition control lengthening out this particular part and it can be differentially elongated.

ATP shows high affinity with Poly(A). Using nearly any RNA strand as template, PAP synthesizes the poly(A)-tail on it. That closed conformation around RNA substrate which allows many adenosine residues to be added by bringing about substantial domain movement within its structure was observed during detailed examination of PAP’s architecture while studying its mechanisms. PIE(polyadenylation inhibition RNA elements), PASF (polyadenylation specificity factors) are among some factors that regulate activity depending on processing and length control for a given poly A tail.
